are you using the asl-menu to navigate your way to Interface Tune CLI ( documented in this man page ) ?

here are my notes for a similar setup:

Yaesu FT-8900	use Carrier From 'usb'; RX level 700

you will probably find it useful to do View COS, CTCSS and PTT Status while kerchunking your interface. don't connect that node to anywhere until the interface settings are settled

if you do not have the radio set to do Tone Squelch, then set CTCSS From to no for now

If the URI PTT LED is lighting up when ASL keys PTT, then ASL is in fact communicating properly with the URI and the URI is pulling the PTT line low on the Mini-DIN-6 output. You can easily confirm that by taking the URI out of the enclosure and then measuring the voltage on the PTT line on the header pads that go to the Mini-DIN-6 jack. (Will be 0V when keyed, ~5V when not). If the radio is then not keying up it could only be because the radio is somehow not configured right, or maybe you have a bad Mini-DIN-6 cable. But usually there's nothing to configure for a Mini-DIN-6 PTT input, that is typically hardwired in radios to key Tx. To confirm it's not the cable you could check the pins on the radio side of the MD6 cable with a DMM and confirm you see < 100Ω between PTT and Gnd when PTT is asserted. If so, ASL, the URI and the cable are doing what they should and the issue would appear to be something in the radio.
