You really haven't provided anything we can troubleshoot with you. If you key up and there's no audio, that's almost always either a problem in simpleusb-tune-menu of having the audio too low or your interface to the soundcard is broken.
The tune menu (option 2 I think) has the little audio meter for RX traffic outbound to allstar (network and other connected nodes). If you see no meter deflection when you are keyed up and speaking into the mic troubleshoot audio path issues from source (normally the node radio) to the sound interface/allstar. My guess is that you dont have any audio there. There are a lot of variables so type of wiring/radio/interface that you are using will help everyone give suggestions.
